BACKGROUND: SPERM DNA DAMAGE IS UNDERLYING AETIOLOGY OF POOR IMPLANTATION AND PREGNANCY RATES BUT ALSO AFFECTS HEALTH OF OFFSPRING AND MAY ALSO RESULT IN DENOVO MUTATIONS IN GERM LINE AND POST FERTILIZATION. THIS MAY RESULT IN COMPLEX DISEASES, POLYGENIC DISORDERS AND CHILDHOOD CANCERS. CHILDHOOD CANCER LIKE RETINOBLASTOMA (RB) IS MORE PREVALENT IN DEVELOPING COUNTRIES AND THE INCIDENCE OF RB HAS INCREASED MORE THAN THREE FOLD IN INDIA IN THE LAST DECADE. RECENT STUDIES HAVE DOCUMENTED INCREASED INCIDENCE OF CANCERS IN CHILDREN BORN TO FATHERS WHO CONSUME ALCOHOL IN EXCESS AND TOBACCO OR WHO WERE CONCEIVED BY ASSISTED CONCEPTION. THE AETIOLOGY OF CHILDHOOD CANCER AND INCREASED DISEASE BURDEN IN THESE CHILDREN IS LIN KED TO OXIDATIVE STRESS (OS) AND OXIDATIVE DNA DAMAGE( ODD) IN SPERM OF THEIR FATHERS. THOUGH SEVERAL ANTIOXIDANTS ARE IN USE TO COMBAT OXIDATIVE STRESS, THE EFFECT OF MAJORITY OF THESE FORMULATIONS ON DNA IS NOT KNOWN. YOGA AND MEDITATION CAUSE SIGNIFICANT DECLINE IN OS AND ODD AND AID IN REGULATING OS LEVELS SUCH THAT REACTIVE OXYGEN SPEUES MEDITATED SIGNAL TRANSDUCTION, GENE EXPRESSION AND SEVERAL OTHER PHYSIOLOGICAL FUNCTIONS ARE NOT DISRUPTED. THUS, THIS STUDY AIMED TO ANALYZE SPERM ODD AS A POSSIBLE ETIOLOGICAL FACTOR IN CHILDHOOD CANCER AND ROLE OF SIMPLE LIFE STYLE INTERVENTIONS LIKE YOGA AND MEDITATION IN SIGNIFICANTLY DECREASING SEMINAL OXIDATIVE STRESS AND OXIDATIVE DNA DAMAGE AND THEREBY DECREASING INCIDENCE OF CHILDHOOD CANCERS. MATERIALS AND METHODS: A TOTAL OF 131 FATHERS OF CHILDREN WITH RB (NON-FAMILIAL SPORADIC HERITABLE) AND 50 CONTROLS (FATHERS OF HEALTHY CHILDREN) WERE RECRUITED AT A TERTIARY CENTER IN INDIA. SPERM PARAMETERS AS PER WHO 2010 GUIDELINES AND REACTIVE OXYGEN SPECIES (ROS), DNA FRAGMENTATION INDEX (DFI), 8-HYDROXY-2'-DEOXY GUANOSINE (8-OHDG) AND TELOMERE LENGTH WERE ESTIMATED AT DAY 0, AND AFTER 3 AND 6 MONTHS OF INTERVENTION. WE ALSO EXAMINED THE COMPLIANCE WITH YOGA AND MEDITATION PRACTICE AND SMOKING STATUS AT EACH FOLLOW-UP. RESULTS: THE SEMINAL MEAN ROS LEVELS (P<0.05), SPERM DFI (P<0.001), 8-OHDG (P<0.01) LEVELS WERE SIGNIFICANTLY HIGHER IN FATHERS OF CHILDREN WITH RB, AS COMPARED TO CONTROLS AND THE RELATIVE MEAN TELOMERE LENGTH IN THE SPERM WAS SHORTER. LEVELS OF ROS WERE SIGNIFICANTLY REDUCED IN TOBACCO USERS (P<0.05) AS WELL AS IN ALCOHOLICS (P<0.05) AFTER INTERVENTION. DFI REDUCED SIGNIFICANTLY (P<0.05) AFTER 6 MONTHS OF YOGA AND MEDITATION PRACTICE IN ALL GROUPS. INCREASED MT DNA MUTATIONS AND ABERRANT REPAIR OF MT AND NUCLEAR DNA DUE TO HIGHLY TRUNCATRED DNA REPAIR MECHANISMS ALL CONTRIBUTE TO SPERM GENOME HYPERMUTABILITY AND PERSISTANT OXIDATIVE DNA DAMAGE. OXIDATIVE STRESS IS ALSO ASSOCIATED WITH GENOME WIDE HYPOMETHYLATION, TELOMERE SHORTENING AND MITOCHONDRIAL DYSFUNCTION LEADING TO GENOME HYPERMUTABILITY AND INSTABILITY. INCREASES IN HIGH-DENSITY LIPOPROTEIN AFTER 120 DAYS WERE NOT SIGNIFICANT (6 MG/DL MEDIAN INCREASE; P=0.15). RESULTS: IN HRV, FREQUENCY DOMAIN ANALYSIS SHOWED A SIGNIFICANT FALL IN LOW FREQUENCY (LF) IN NORMALIZED UNITS (NU) AND LF: HIGH FREQUENCY (HF) RATIO AND SIGNIFICANT RISE IN HF IN NU IN THE YOGA GROUP (DEPICTING PARASYMPATHETIC DOMINANCE) AGAINST A SIGNIFICANT RISE IN LF (NU) AND LF: HF RATIO AND SIGNIFICANT FALL IN HF (NU) IN NON-YOGA GROUP (INDICATING SYMPATHETIC DOMINANCE). TIME DOMAIN ANALYSIS SHOWED A SIGNIFICANT DECREASE IN STANDARD DEVIATION OF NN INTERVALS IN NON-YOGA GROUP AGAINST NONSIGNIFICANT CHANGES IN YOGA GROUP INDICATING DETERIORATION IN PARASYMPATHETIC ACTIVITY IN NON-YOGA GROUP. A TOTAL OF 55 WOMEN AND 38 MEN PARTICIPATED IN THE STUDY. DIFFERENCES BETWEEN MEN AND WOMEN AT ENROLLMENT INCLUDED: WOMEN REPORTED SIGNIFICANTLY GREATER WITHDRAWAL (P<0.005), ANXIETY (P=0.032), AND DEPRESSION (P=0.027) SYMPTOMS THAN MEN. MORE WOMEN THAN MEN (91% VS. 66%) REPORTED HAVING BEEN TOLD BY THEIR DOCTOR TO QUIT SMOKING (P=0.003), HAD AN EXISTING SMOKING-RELATED ILLNESS (33% VS. 13%; P=0.032), AND REPORTED SMOKING FOR WEIGHT CONTROL (15% VS. 0%; P=0.014). RESULTS SHOWED GOOD FEASIBILITY FOR RECRUITING BOTH MEN AND WOMEN INTO A STUDY USING YOGA AS A COMPLEMENTARY THERAPY FOR SMOKING CESSATION. BACKGROUND: OBESE PERSONS OFTEN FIND PHYSICAL ACTIVITY DIFFICULT. THE EFFECTS OF A YOGA AND DIET CHANGE PROGRAM, EMPHASIZING BREATHING TECHNIQUES PRACTICED WHILE SEATED, WAS ASSESSED IN OBESE PERSONS. MATERIAL/METHODS: A SINGLE GROUP OF 47 PERSONS WERE ASSESSED ON THE FIRST AND LAST DAY OF A YOGA AND DIET CHANGE PROGRAM, WITH 6 DAYS OF THE INTERVENTION BETWEEN ASSESSMENTS. THE ASSESSMENTS WERE: BODY MASS INDEX (BMI), WAIST AND HIP CIRCUMFERENCES, MID-ARM CIRCUMFERENCE, BODY COMPOSITION, HAND GRIP STRENGTH, POSTURAL STABILITY, SERUM LIPID PROFILE AND FASTING SERUM LEPTIN LEVELS. PARTICIPANTS PRACTICED YOGA FOR 5 HOURS EVERY DAY AND HAD A LOW FAT, HIGH FIBER, VEGETARIAN DIET. LAST AND FIRST DAY DATA WERE COMPARED USING A T-TEST FOR PAIRED DATA. RESULTS: FOLLOWING THE 6-DAY RESIDENTIAL PROGRAM, PARTICIPANTS SHOWED A DECREASE IN BMI (1.6 PERCENT), WAIST AND HIP CIRCUMFERENCES, FAT-FREE MASS, TOTAL CHOLESTEROL (7.7 PERCENT DECREASE), HIGH DENSITY LIPOPROTEIN (HDL) CHOLESTEROL (8.7 PERCENT DECREASE), FASTING SERUM LEPTIN LEVELS (44.2 PERCENT DECREASE) AND AN INCREASE IN POSTURAL STABILITY AND HAND GRIP STRENGTH (P<0.05, ALL COMPARISONS). CONCLUSIONS: A 6-DAY YOGA AND DIET CHANGE PROGRAM DECREASED THE BMI AND THE FAT-FREE MASS. TOTAL CHOLESTEROL ALSO DECREASED DUE TO REDUCED HDL LEVELS. PURPOSE: TO OBTAIN ESTIMATES OF TIME TO RECRUIT THE STUDY SAMPLE, RETENTION, FACILITY-BASED CLASS ATTENDANCE AND HOME PRACTICE FOR A STUDY OF YOGA IN BREAST CANCER SURVIVORS, AND ITS EFFICACY ON FATIGUE, QUALITY OF LIFE (QOL), AND WEIGHT CHANGE. METHODS: SIXTY-THREE POST-TREATMENT STAGES 0-III BORDERLINE OVERWEIGHT AND OBESE (BODY MASS INDEX >/= 24 KG/M(2)) BREAST CANCER SURVIVORS WERE RANDOMLY ASSIGNED TO A 6-MONTH, FACILITY- AND HOME-BASED VINIYOGA INTERVENTION (N = 32) OR A WAITLIST CONTROL GROUP (N = 31). THE YOGA GOAL WAS FIVE PRACTICES PER WEEK. PRIMARY OUTCOME MEASURES WERE CHANGES IN QOL, FATIGUE, AND WEIGHT FROM BASELINE TO 6 MONTHS. METHODS: WE CONDUCTED A CONTROLLED TRIAL WITHIN AN UNIVERSITY-AFFILIATED HOSPITAL. 173 CHINESE MEN AND WOMEN AGED 18 OR ABOVE WERE ASSIGNED TO EITHER THE YOGA INTERVENTION GROUP (N = 87) OR THE CONTROL GROUP (N = 86). PRIMARY OUTCOMES INCLUDED 12-WEEK CHANGE IN METABOLIC RISK FACTORS AND METS Z SCORE. SECONDARY OUTCOME WAS HRQOL (MEDICAL OUTCOMES SHORT FORM SURVEY AT 12 WEEKS). RESULTS: THE MEAN AGE OF PARTICIPANTS WAS 52.0 (SD 7.4, RANGE 31-71) YEARS. ANALYSIS INVOLVING THE ENTIRE STUDY POPULATION REVEALED THAT THE YOGA GROUP ACHIEVED GREATER DECLINE IN WAIST CIRCUMFERENCE (P<0.001), FASTING GLUCOSE (P<0.01), TRIGLYCERIDES (P<0.05), AND METS Z SCORE (P<0.01).
