BACKGROUND: SEPSIS IS AN INCREASINGLY SIGNIFICANT CHALLENGE THROUGHOUT THE WORLD AS ONE OF THE MAJOR CAUSES OF PATIENT MORBIDITY AND MORTALITY. CENTRAL TO THE HOST IMMUNOLOGIC RESPONSE TO SEPSIS IS THE INCREASE IN CIRCULATING MYELOID-DERIVED SUPPRESSOR CELLS (MDSCS), WHICH HAVE BEEN DEMONSTRATED TO BE PRESENT AND INDEPENDENTLY ASSOCIATED WITH POOR LONG-TERM CLINICAL OUTCOMES. MDSCS ARE PLASTIC CELLS AND POTENTIALLY MODIFIABLE, PARTICULARLY THROUGH EPIGENETIC INTERVENTIONS. IN THIS STUDY, WE EVALUATED THE EFFECTS OF A COMBINATION OF NUTRACEUTICAL SUPPLEMENTS (NS) ON TELOMERE LENGTH (TL) IN HEALTHY VOLUNTEERS WITH NO MEDICAL HISTORY OF ANY DISEASE. WE ESTIMATED THE LENGTH OF SINGLE TELOMERES IN METAPHASE SPREAD LEUKOCYTES, ISOLATED FROM PERIPHERAL BLOOD, USING QUANTITATIVE?FLUORESCENT IN SITU HYBRIDIZATION (Q?FISH) ANALYSIS. THE LENGTH OF THE WHOLE TELOMERE GENOME WAS SIGNIFICANTLY INCREASED (P<0.05) FOR THE MEAN, 1ST QUARTILE AND MEDIAN MEASUREMENTS IN THE EXPERIMENTAL GROUP. SIMILAR FINDINGS WERE OBSERVED FOR SHORT TL (20TH PERCENTILE) (P<0.05) FOR THE MEDIAN AND 3RD QUARTILE MEASUREMENTS IN THE NS GROUP, COMPARED TO THE CONTROL GROUP. THE BENEFICIAL EFFECTS OF THE SUPPLEMENTS ON THE LENGTH OF SHORT TELOMERES REMAINED SIGNIFICANT (P<0.05) FOLLOWING ADJUSTMENT FOR AGE AND SEX. TELOMERES WERE MODERATELY LONGER IN FEMALE PATIENTS COMPARED TO THE MALE PATIENTS. BECAUSE OF HETEROGENEITY OF TUMORS, USUALLY MULTIPLE MARKERS DISTRIBUTED THROUGHOUT THE HUMAN GENOME NEED TO BE ANALYZED. THIS IS LABOR INTENSIVE AND DOES NOT ALLOW FOR HIGH THROUGH-PUT SCREENING. IN THIS STUDY, WE INVESTIGATE THE PRIMING EFFECTS OF URIC ACID EXPOSURE ON THE CYTOKINE PRODUCTION OF PRIMARY HUMAN CELLS UPON STIMULATION WITH GOUT-RELATED STIMULI. CYTOKINE PRODUCTION WAS MEASURED BY ELISA; MRNA LEVELS WERE ASSESSED USING QPCR. RESULTS: THE PRODUCTION OF INTERLEUKIN (IL)-1BETA AND IL-6 WAS HIGHER IN PATIENTS COMPARED WITH CONTROLS AND THIS CORRELATED WITH SERUM URATE LEVELS. THIS STUDY EXPLORED THE ASSOCIATION OF DNAM AGE OR AA WITH PSYCHONEUROLOGICAL (PN) SYMPTOMS, INCLUDING COGNITIVE IMPAIRMENT, FATIGUE, SLEEP DISTURBANCES, PAIN, AND DEPRESSIVE SYMPTOMS, IN BREAST CANCER SURVIVORS OVER A 2-YEAR PERIOD. CONCLUSION: OUR FINDINGS SUGGEST DNAM AGE AND AA MAY BE ASSOCIATED WITH PN SYMPTOMS OVER THE COURSE OF CANCER TREATMENT AND SURVIVORSHIP. AS DEMETHYLATION FACTORS, TEN-ELEVEN TRANSLOCATION (TET) FAMILY MEMBERS HAVE BECOME A FOCUS IN RECENT STUDIES OF NEUROLOGICAL DISORDERS. HERE, WE QUANTIFIED AND LOCALIZED TET1, TET2 AND 5-HYDROXYMETHYLCYTOSINE (5-HMC) IN THE TEMPORAL LOBE CORTEX OF DRE PATIENTS (N = 27) AND TRAUMATIC BRAIN HEMORRHAGE CONTROLS (N = 10) BY IMMUNOCHEMICAL STAINING. TET2 AND ATP BINDING CASSETTE SUBFAMILY B MEMBER 1 (ABCB1) EXPRESSION PATTERNS WERE DETERMINED IN THE ISOLATED BRAIN CAPILLARIES OF DRE PATIENTS.
